What Alpha-Ketoglutarate Is—and Whether It's Safe for Aging Dogs

An 11-year-old, 62-pound Labrador retriever comes in for her annual senior workup. Bloodwork is unremarkable, her joints are stiff but manageable, and her owner has a browser tab open to a supplement claiming to “reverse aging” with alpha-ketoglutarate. He wants to know, plainly, whether to add it to her bowl. That question—not the marketing copy—is the one worth answering carefully.

What alpha-ketoglutarate actually is

Alpha-ketoglutarate (AKG) is not an exotic import. It’s a metabolite your dog’s cells already make and use every day, generated in the Krebs cycle as cells convert nutrients into usable energy. It also plays a role in amino acid metabolism and collagen synthesis. That endogenous status is exactly why the safety conversation is more nuanced than “new ingredient, unknown risk”—the body isn’t meeting AKG for the first time. What’s new is the idea of supplementing it in a calcium-buffered form, at doses well above what normal metabolism produces, to see whether that shifts the trajectory of aging itself.

What the mouse data actually shows

The foundational study here comes from researchers at the Buck Institute, published in Cell Metabolism. In aged mice given calcium alpha-ketoglutarate (Ca-AKG), the treated group lived longer and, notably, spent more of that extra time in good health rather than declining—what the Cell Metabolism / PubMed: Cell Metabolism paper frames as a compression of morbidity rather than a simple lifespan bump. The proposed mechanism runs through reduced systemic inflammation, including a drop in inflammatory cytokines. That’s a genuinely interesting result. It is also a mouse result, run at a specific dose in a specific strain, and mouse metabolism is not dog metabolism laid over a different-shaped body.

I once worked through a similar gap with an owner who’d already started her 9-year-old terrier mix on a human NAD+ precursor product, reasoning that “cellular support” was cellular support regardless of species or dose. Six weeks in, nothing had visibly changed except her confidence that it was working. That’s a meaningfully different evidence tier than a mouse study—closer to what regulators and clinicians want to see—but it still isn’t a dog study, and biological-age biomarkers in people don’t automatically translate into a validated readout for canine aging.

The closest thing to a dog-specific answer

The most direct canine longevity work underway right now doesn’t actually involve AKG as an over-the-counter supplement—it involves a related compound in a conditionally regulated drug candidate. Loyal, a veterinary biotech company, is running the STAY trial, a large, blinded, placebo-controlled study enrolling roughly 1,000 to 1,300 senior dogs across dozens of practices, evaluating a drug intended to extend healthy lifespan by targeting age-related metabolic decline. It’s being developed for dogs age 10 and up weighing at least 14 pounds, administered as a prescribed daily pill rather than a shelf supplement. That distinction matters: a compound moving through veterinary drug development, with Target Animal Safety data submitted to the FDA, sits on different regulatory footing than an ingredient added to a chew because a mouse paper looked promising.

Here’s where the edge case shows up in practice. A 13-year-old, 48-pound mixed-breed dog with mild chronic kidney disease is exactly the patient where “AKG is endogenous, so it’s inherently safe” stops being a sufficient answer—amino acid and nitrogen metabolism intersect with kidney function, and a dog with reduced renal reserve is not the same physiological system as a healthy laboratory mouse. By the three-week mark on any new longevity supplement in a dog like this, I want a recheck of kidney values, not just an owner’s impression that her coat looks shinier. The mechanism that looks clean in a healthy model can behave differently once an organ system is already compromised, which is exactly why species- and case-specific data, not extrapolation, has to drive the decision.

Where that leaves an owner today

For a healthy senior dog, the biological plausibility of AKG is real: it’s an endogenous metabolite with a published mouse longevity study and human trials now underway to test the same mechanism. What doesn’t exist yet is a published, dog-specific safety or efficacy trial for AKG as a supplement at a defined canine dose. That gap is worth sitting with rather than talking around. Until it closes, I tell owners to treat AKG the way we treat most emerging longevity compounds in dogs: promising signal, not yet a settled answer, and not a substitute for the basics that already have decades of veterinary evidence behind them—appropriate joint support, weight management, and routine senior bloodwork. If your dog has kidney disease, is on other supplements, or is already showing cognitive or mobility changes, that’s a conversation for your veterinarian before it’s a purchase decision.
